The detailed physicochemical characterization and CD complexation of four key TKIs—erlotinib, gefitinib, vandetanib, and lapatinib—were investigated. A broad set of analytical techniques was applied to study the acid–base properties, lipophilicity, and solubility of these drugs, highlighting low solubility as a fundamental barrier to their bioavailability.
CD complexation emerged as a promising strategy to improve the performance of the studied TKIs. The stoichiometry and stability constants of inclusion complexes formed with native CDs (α-CD, β-CD, γ-CD) and substituted derivatives were characterized using different analytical methods.
Phase-solubility studies demonstrated that CD complexation can substantially enhance the water solubility of the investigated TKIs, with the potential to improve bioavailability. In addition, NMR and molecular modelling were used to examine the stoichiometry and geometry of the complexes, providing mechanistic insights that can support formulation design and inform the further development of TKI-based therapeutics.
